Abstract

The association of (. ) infection and parasitic infections including (), especially in childhood, is widely recognized to be high in developing communities. We aimed to study the impact of concomitant intestinal parasitic and infections on the different clinical presentation of infected children and whether this coinfection could in turn cause any alteration in the clinical manifestations of each other. This cross-sectional study included 150 children of both sexes with their age ranging from 1 to 15 years, having gastrointestinal complaints, throughout 8 months duration. All cases were subjected to full history taking, clinical examination and stool analysis by direct wet smear and formalin-ethyl acetate concentration technique, permanent staining with cold acid fast stain in addition to coproantigen detection in stool. Parasitic infection was recorded in 58.6% of patients, with the most detected parasite (35.2%). Cases infected with were 63 cases (42%) of which 61.9% of cases showed associated parasitic infection. Diarrhea was the most frequent complaint (63.2%) in cases infected with intestinal parasites, while it was less frequently recorded in co-infected cases (35.8%) and in cases with infection only (29.1%) ( value 0.0008). On the other hand, vomiting was less recorded in coinfected cases than cases with infection. Coinfection with intestinal parasites (including ) and . could modulate the clinical manifestation of each other especially diarrhea in parasitic infections and vomiting in infection.

Abstract

The influence of and variants on colorectal cancer (CRC) susceptibility and their impact on //epithelial-mesenchymal transition (EMT) and //EMT axes in CRC are unknown. We investigated the influence of and on the risk of CRC and adenomatous polyps (AP), their impact on the long noncoding RNAs and expression and their target , /E-cadherin pathways, along with their potential as early CRC biomarkers. Overall, 280 individuals were recruited: 140 patients with CRC, 40 patients with AP, and 100 healthy volunteers. Genotyping and serum expression profiles were assessed using qPCR. The EMT biomarker, E-cadherin, was measured by ELISA. was associated with increased CRC risk, whereas was protective. Serum and were upregulated in CRC and AP patients versus healthy controls, whereas, , and E-cadherin were downregulated in CRC versus non-CRC groups. showed superior diagnostic potential for CRC and predicted CRC risk among non-CRC groups in the multivariate logistic analysis. , , and levels were correlated with E-cadherin, tumor stage, lymph node and distant metastasis. E-cadherin was lost in metastatic vs. non-metastatic CRC. genotype carriers showed higher E-cadherin levels than carriers. was correlated with lymph node status. For the first time, and are potential genetic CRC predisposition markers, with possibly impacting the EMT process. Serum , , and are novel non-invasive diagnostic biomarkers that could improve the clinical outcome of CRC.

Abstract

BACKGROUND: Systemic lupus erythematosus (SLE) is a systemic autoimmune disease. The autoimmune regulator () is a master regulator of self-tolerance development. mutations lead to the development of autoimmune polyglandular syndrome type 1 while polymorphisms have been linked to organ-specific autoimmunity. The study is aimed at addressing the association between polymorphisms, rs2075876 (G > A) and rs760426 (A > G), and SLE susceptibility and expression in Egyptian patients.

METHODS: Ninety-nine patients were included. One hundred and ten, and 123 control subjects were genotyped for rs2075876 and rs760426, respectively. Lupus severity was assessed using the Lupus Severity of Disease Index and Lupus Severity Index (LSI). Systemic Lupus International Collaborating Clinics (SLICC)/American College of Rheumatology (ACR) damage index was considered. Genotyping was done using StepOne Real-Time PCR. rs760426 GG was more frequent in the patients under the genotype level (14.1% vs. 4.9%, = 0.032) and recessive model (14.1% vs. 4.9%, = 0.017, OR = 3.2 (1.2-8.7)). Musculoskeletal involvement and nephritis were associated with rs2075876 under the dominant (97.9% vs. 80.8%, = 0.009, OR = 11 (1.3-89.2)) and recessive models (100% vs. 69.3%, = 0.032), respectively; and both were linked to rs2075876 at the allelic level: 98.3% vs. 85%, = 0.005, OR = 10.1 (1.3-76.6) and 82.8% vs. 68.6, = 0.041, OR = 2.2 (1-4.7), respectively. Patients with rs2075876 A alleles had a higher damage index ( 1 ± 1.3 vs. 0.6 ± 1.1, = 0.045) while the LSI was greater in patients with rs2075876 (8.5 ± 0.5 vs. 7.8 ± 1.3, = 0.002) and rs760426 (8.6 ± 11 vs. 7.8 ± 1.2, = 0.031) under the recessive models. rs760426 could share in SLE susceptibility while rs2075876 could influence the disease expression and burden in Egyptian patients.
