Abstract

New 1-pyrazolo[3,4-d]pyrimidine derivatives were designed and synthesised to act as epidermal growth factor receptor inhibitors (EGFRIs). The synthesised derivatives were assessed for their anti-proliferative activities against A549 and HCT-116 cancer cells. Compounds and showed potent anti-proliferative activities. Compound was the most promising member with IC values of 8.21 and 19.56 µM against A549 and HCT-116, respectively. Compounds and were evaluated for their kinase inhibitory activities against wild EGFR (EGFR). Compound was the most potent member showing an IC value of 0.016 µM. In addition, compound showed noticeable activity against mutant EGFR (EGFR) (IC = 0.236 µM). Flow cytometric analyses revealed that compound is a good apoptotic inducer and can arrest the cell cycle at S and G2/M phases. Furthermore, it produced an 8.8-fold increase in BAX/Bcl-2 ratio. Molecular docking studies were carried out against EGFR and EGFR.

A simple and efficient approach for profiling the greenness of high performance liquid chromatography (HPLC) methods is presented. This environmental assessment tool (EAT) takes into consideration the environmental, health and safety issues for all solvents involved in the chromatographic method, and calculates a total score that can be used for comparison of the greenness of different methods. A software, HPLC-EAT, has been designed to facilitate the calculation and can be downloaded free of charge at HPLC-EAT was successfully applied for a set of different HPLC methods from the literature, including both analytical and preparative chromatography. The performance of the tool was validated and it was further combined with another free software Eco-solvent tool to perform life cycle assessments of waste disposal options of distillation or incineration. HPLC-EAT can be routinely used in method development to calculate the greenness beside the conventional standards of accuracy, robustness and reproducibility.

In serial multi-stage manufacturing systems (SMMS), optimization of part quality inspection planning (PQIP), buffer allocation problem (BAP), and preventive maintenance (PM), individually and jointly, is attracting researchers’ attention. The model formulation for complicated manufacturing systems and the previously mentioned joint decisions is very beneficial given the interdependencies between the various manufacturing functions. As a result, this paper evaluates the literature on joint optimization of the multi-stage serial production system. The literature is classified based on the decision variables basis to represent each manufacturing function [inspection sample size and allocation (PQIP), buffer sizing and allocation (BAP), and preventive maintenance scheduling (PM)], and a general example model is presented in each classification, with a summary of recent studies, solution methods, research gaps, and future research recommendations. In the integrated models, almost all the studies considered only two functions, with that it is worth noting that research into the optimization of over two functions is still in its beginning. Furthermore, most studies neglected many of the real industrial settings that should also be integrated into the model. And finally, there was no specific solution technique recommended in the literature, yet a general simulation optimization method was used to generate and evaluate the combinatorial complex joint models.
