In the present study, the nucleocapsid protein and the two subunits of spike protein (S1 and S2) of bovine coronavirus (Mebus strain) were individually expressed in Spodopetra frugiperda (Sf9) insect cells using recombinant baculoviruses. The processes of generation of the recombinant baculoviruses carrying the target genes and characterization of the expressed proteins were conducted as follow:
1- Bovine coronavirus (Mebus strain) was propagated for four successive passages in MDBK cells. The fourth passage was titrated and revealed a virus titer of 105 TCID50/ml.
2- The propagated virus was characterized using plate haemagglutination test (utilizing rat and adult chicken RBCs) and indirect fluorescent antibody technique.
3- BCV-RNA was extracted from the infected cells and coding sequence of the target genes was amplified by RT-PCR.
4- The RT-PCR products were cloned in the baculovirus transfer vector [pBlueBac4.5/V5-His-TOPO®].
5- Positive recombinant plasmids carrying the target genes in correct orientation were identified for each gene insert and selected for the plasmid extraction and transfection process.
6- The cloned genes were introduced in the genome of Autographa californica nuclear polyhydrosis virus (AcMNPV) under control of the polyhedrin promoter, through a process of homologous recombination between the transfer vector (recombinant plasmid) and a linearized replication-defective baculovirus DNA (Bac-N-Blue™).
7- Recombinant baculoviruses were selected using plaque purification technique and used for generation of P-1 virus stocks.
8- After PCR analysis of the P-1 virus stock, the pure recombinant viruses were propagated to generate high-titer (P-2) viral stocks. The propagated viruses were titrated using plaque assay. Recombinant viruses with satisfactory titers were used for the in vitro expression studies.
9- High-level expression of the target proteins was indicated by the indirect immuno-fluoresent test and solid phase ELISA.
10- Application of western blot assay for characterization of the expressed proteins revealed detection of N protein in cellular extracts of infected cells under both reducing and non-reducing conditions, while S1 and S2 subunit proteins were not recognized in both conditions.

The use of such proteins in the field of diagnosis and vaccine development will aid in controlling the virus-induced disease problems in Egypt. Moreover, the baculovirus expression system will provide a magic tool for production of a huge range of recombinant proteins that may be invested in many aspects of science and technology.

Abstract

This study demonstrated a significant delay in sensory and motor block regression, in each of the perineural and intravenous dexamethasone groups, and also in the perineural compared to the intravenous dexamethasone group. Duration of analgesia was significantly prolonged in the perineural dexamethasone group, compared to the intravenous dexamethasone and control groups, and also in the intravenous dexamethasone group, compared to the control group. Postoperative pain scores were significantly lower in the perineural dexamethasone group.
